傅里叶红外光谱仪(Fourier Transform Infrared Spectroscopy,简称FTIR)是一种非常重要的化学分析工具。它可以通过检测样品中特定波长的红外辐射,获取有关该样品化学组成、结构和功能性的信息。

FTIR原理与分类

FTIR基于傅里叶变换将信号转换为频域图像进行测试。当我们向样品施加太阳光等强度不同的电磁辐射时,它会吸收频率相对应的能量并产生振动。

因此,可以使用一个干涉计来记录被吸收之后未被普通太阳光线反射或透过去的剩余电磁辐射,并将这些数据进行处理以获得三个主要类型:透射式(Transmission),反射式(Reflection)和总反射式几何(Attenuated Total Reflectance, ATR)型。其中最流行且广泛使用者为ATR型。

ATR技术优势与机理

ATT技术可以解决传统FTIR技术中的大多数问题。在透射模式下,必须处理较小量且需要整理为非液态物质进行测试;而反射模式则通常利用ZnSe晶体来实现测定固体样品。

相比之下,ATR模式由于直接与样品接触并使用内部反射原理记录信号外还具有以下好处:

  • 可针对不同类型的样本进行快速分析和测量
  • 无需任何特殊准备步骤(如破碎或粉碎)即可以较少的化学修改操作直接获得数据
  • デetect sample components at low concentrations and without cross-contamination between samples, making it suitable for a range of industries including pharmaceuticals, food technology, manufacturing and engineering.

ATR技术应用

Numerous domains have adopted ATR technology to conduct accurate infrared spectroscopic analyses. These include but are not limited to:

  • Hazard detection:Infrared spectra can be used to easily identify hazardous substances in materials that come into contact with humans or the environment.
  • Biochemical studies: bacteriology (e.g. distinguishing bacterial strains), protein dynamics research (such as structural characterization) or medical diagnostics (for example detecting viral infections or diseases).
  • Materials analysis: AT can be used to identify and characterize materials in the construction, engineering and manufacturing industries.
  • Pharmaceutics: Infrared spectral technology is a proven method for ensuring product quality controls in drug production processes. This is particularly true for testing APIs (active pharmaceutical ingredients) without having to prepare them by changing their molecular structure, which could change their efficacy.

Much of the literature surrounding ATR infrared spectroscopy has focused on its application within existing domains where it has proved successful.
